Rodeo champ Gene Autry inherits half interest in both a ranch and a mine that provides steady employment for the surrounding rancheros. Unfortunately, the other half goes to Easterner Barbara Erwin (Carol Hughes), who is only interested in monetary remuneration. To convince Gene to buy her share, Barbara enters into an unholy alliance with unscrupulous attorneys Arnold (Ivan Miller) and Fry (Sam Flint), who, without their client's consent, hire a gang of thugs headed by Tommick (John Merton). A piece eventually breaks off, sending both of them into a boulder field. They play hide-and-seek a while. Julius then takes off his fur and sends it out as a decoy while he sneaks up behind the bad guy with a club and beats him into the ground. Alice comes up to thank him; ashamed by his nakedness, he hides behind a rock and puts his fur back on, then accepts her thanks.

Like most serials in the '50s, Riding with Buffalo Bill consisted of quite a bit of budget-stretching stock footage telling a highly fictionalized account of Buffalo Bill Cody aiding a group of ranchers in their defeat of a local crime lord. The serial's assistant director, Leonard Katzman, later produced the long-running television series Gunsmoke and Dallas.
